"I'm really excited about what the team can achieve next season after the huge strides we made this year," said Vos.
Meanwhile, lock Olly Kohn, 25, has joined Quins after being released by Bristol following a career plagued by injuries at the Memorial Stadium.
Vos, the former South Africa captain, has led Quins to 22 victories from 23 league matches this term.

And he is set to skipper the side against Bedford in the final of the Powergen National Trophy at Twickenham on Sunday.
"We've had to toughen up as a squad and the guys have stuck to their guns and battled extremely well to get us back in the Premiership," he said.

"One of our objectives for this year was to win the double and we've given ourselves that opportunity thanks to a lot of hard work."
In addition to Vos, Quins have retained the services of many of their academy players, including Mike Brown, Alex Rogers, Chris Robshaw and Tosh Masson, who were all part of the England Under 21 squad that won the Grand Slam in this years Six Nations.
And Jordan Turner-Hall, who is in the England Under-19 squad competing in the IRB Under-19 World Championship in Dubai this month, has also committed to the club for another two years.
"One of the most pleasing aspects of this season has been the performances of the academy players playing for the first XV," said Quins boss Dean Richards.
"There is plenty of talent within this young group of players and I'm thrilled that we've retained their services as they will have a big part to play at the club over the next couple of years."
