Doncaster striker Billy Sharp, who is on a season-long loan from Sheffield United, scored the only goal to beat Sheffield Wednesday.
His 78th-minute header from Simon Gillett's cross was Sharp's sixth goal in as many games.
Leon Clarke went close for Wednesday before the break but shot wide.
Later the hosts' James O'Connor headed over from a corner before Sharp struck. There was still time for a chance for Mark Beevers but he volleyed just over.
Doncaster manager Sean O'Driscoll:
"It was a very dour game. They came to do a job, tried to stop us from playing and credit to them.
"On overall play, if we'd have walked off with a draw I would have said 'OK'.
"We worked hard for it, it wasn't one of our better performances, but they didn't allow us to play so you've got to give them credit for that."
Sheffield Wednesday boss Brian Laws:
"I got what I asked for. That was a derby-like performance. We were dogged, resilient and committed.
"The commitment was 100% and I've said to the players it doesn't matter what happens in terms of games, if you put that effort in we'll turn it round.
"We've got to believe. The fans believe we can do it, they were outstanding."
