Boeta Dippenaar warmed up for the triangular against England and Zimbabwe with an aggressive unbeaten century.
And South Africa's bowlers showed they were in good heart as they bowled Sussex out in the 31st over, with Charl Langeveldt and Paul Adams taking three wickets each.
Dippenaar was not in the orginal squad for the one-day phase of South Africa's tour, but was called up because of Neil McKenzie's back problem.
Having made 76 in Ireland on Wednesday, he followed up with 101 not out off 90 balls, with two sixes and seven fours.
Robin Martin-Jenkins was the pick of the home side's attack, sending down 10 overs at a cost of only 34 runs, and he was rewarded with the wickets of Herschelle Gibbs and Andrew Hall.
South Africa's innings was launched with a stand of 66 between Gibbs and captain Graeme Smith, who was first to go for 31 when he was caught off the bowling of Carl Hopkinson.

Gibbs was lbw for the same score and Jacques Rudolph followed for 19 when wicket-keeper Matt Prior took an athletic catch off the bowling of Pakistani leg-spinner Mushtaq Ahmed.
Martin van Jaarsveld helped Dippenaar put on 82 for the fourth wicket before he was lbw to Kevin Innes for 46.
Mark Boucher, Shaun Pollock and Hall fell cheaply over the closing overs but Dippenaar tore into the home attack and reached three figures just before the end of their overs.
Sussex's reply started fairly as Richard Montgomerie and Michael Yardy put on 27 runs for the first wicket.
But Langeveldt's breakthrough to remove Yardy for 12 opened the floodgates for a South African attack.
The hosts lost 7-45 as the gulf between the two sides became apparent, and it was largely thanks to skipper Chris Adams' 32 that triple figures were reached.
Andre Adams took two late wickets before Makhaya Ntini added to his earlier dismissal of Montgomerie to end the match with the wicket of Jason Lewry.
